I have an interview with a Yale alum tomorrow... totally unexpected... he only called yesterday, rather short notice. What do you think is the most important thing to do/convey during an interview (job, college, etc) to ace it?

If you can answer both of those questions fully, you can answer anything you will be asked. Don't give him a rehearsed speech. Have a conversation with him and tell him what you want him to know. Ask specific, pointed questions to better understand what Yale has to offer. Justin is right; let him know that you are confident of your abilities without sayng so.
One word of caution - When you have said what you need to say, when you have answered the questions asked of you, stop talking. Interviewees almost always talk too much. That shows that they are nervous. You have no reason to be nervous, so you have no reason to chatter away.
IF, I have read almost all of your posts here at AnswerPool. You are most certainly what Yale wants. Keep that in mind.
