The general rule for home systems is to shut off the computer if you're leaving the house or sleeping, due to the (slight) fire hazard. Otherwise, once the machine is on, leave it on for the day.
It's a good idea to allow power management to put the monitor into standby ("sleep mode") when there has been no activity for some time (say, 20 minutes).
Running a screensaver or displaying a blank screen still results in normal power consumption, which standy mode greatly reduces. For example, my monitor draws about 130 watts normally, but only 8 watts in standby.
If your computer consistently wakes from standby mode, you can place the machine in standby when not in use. There are a lot of hardware combinations which do not deal well with standby, however, and if the system ever fails to wake from sleep mode, don't use it again.
Leaving the system on 24 hours a day, seven days a week, does no harm at all. Windows should still be periodically restarted, however, at least once a day for Windows 95,98, or ME, and about once a week for Windows NT, 2000, or XP.
